The first settlers reached what is now Westbury in 1658, on the Hempstead Plains, and many were Quakers. Richard Stites and his family built a homestead that year, and an English Quaker named Edmond Titus later arrived with his son Samuel. In 1675 Henry Willis, another English Quaker, named the area Westbury after his hometown, Westbury in Wiltshire. The first Society of Friends meeting house went up in 1700.
Many freed Black residents built their own homesteads on open land here. In 1834, with help from Quakers, the freed families and their descendants constructed the New Light Baptist Church, which became Westbury A.M.E. Zion Church and stands at Union Avenue and Grand Boulevard. Black families also arrived along the Underground Railroad, and some stayed after finding shelter in secret rooms in Quaker homes. After the Hessians settled to the east, the area they chose took the name New Cassel.

Jericho Turnpike began as a trail used by the Massapequa and later divided the early homesteads to the north from the Hempstead Plains to the south. It now connects Westbury with Mineola, Syosset and the Long Island Expressway. Post Avenue grew into a commerce center for the surrounding estates and received electricity in 1902.
The Long Island Rail Road built through Westbury by 1837, and Westbury station on the Main Line is 0.4 miles from the village. The Northern State Parkway was built through the area in 1938. Westbury recorded 15,404 residents in 2020.
